KARDIA-1: one zilebesiran shot lowers 24-hour systolic pressure by up to 16.7 mmHg for months

Alnylam PharmaceuticalsZilebesiran

The KARDIA-1 Phase 2 trial in 378 people with mild-to-moderate hypertension found that a single injection of zilebesiran cut 24-hour systolic blood pressure by 14.1 to 16.7 mmHg more than placebo at three months. Office pressure was still clearly lower at six months, and blood levels of the target protein angiotensinogen fell by 88 to 98%.

  • 378 patients, randomised to placebo or one of several dose schedules.
  • 24-hour systolic pressure fell 14.1-16.7 mmHg vs placebo at 3 months.
  • Office systolic pressure was 7.5-12.1 mmHg lower than placebo at 6 months.
  • Angiotensinogen fell 88-98% depending on dose, and stayed down through six months.
