Intellia says enrollment has restarted in both nex-z Phase 3 trials

Intellia has restarted enrollment in both Phase 3 trials of nex-z, its one-time CRISPR therapy that switches off the TTR gene in the liver to treat ATTR amyloidosis. The trials were halted in late 2025 under an FDA clinical hold after rare, severe liver-enzyme rises. A genetic analysis of more than 600 patient samples found the worst cases clustered in carriers of one immune-gene (HLA) variant, which gives doctors a way to screen for risk.

  • MAGNITUDE tests nex-z in ATTR cardiomyopathy (heart); MAGNITUDE-2 in hereditary ATTR with polyneuropathy (nerves).
  • Investigators and patients enrolled or entering screening will get their HLA genotype results.
  • Intellia expects MAGNITUDE-2 enrollment to finish in the second half of 2026.
  • Cash was $628.4 million at 30 June 2026, which Intellia says funds operations at least into 2028.
