Technical approach
Binding and blocking mRNA
ASOs are modified DNA/RNA that pair with target mRNA, causing cells to destroy it before protein translation.
Reducing TG inhibition
Olezarsen targets APOC3 mRNA to block the protein that inhibits triglyceride breakdown.
Longer-lasting than siRNA
ASOs trigger RNase H, which degrades the target RNA; they thus have longer duration of action than siRNA, allowing monthly rather than quarterly dosing.
