ESO-T01 Phase 1 for multiple myeloma published in Nature Medicine; all patients developed Grade 3+ cytokine release syndrome

Results from the Phase 1 trial of ESO-T01, an in vivo CAR-T therapy for relapsed/refractory multiple myeloma, were published in Nature Medicine. The trial enrolled 5 heavily pretreated patients. All developed Grade 3+ cytokine release syndrome within 24 hours, attributed to immune response to the lentiviral vector. Transient cytopenias and reversible liver enzyme elevations were common.

  • 4 of 5 patients achieved objective responses; 3 stringent complete remissions
