  • One disease at a timeEach aging drug must first work in one narrow disease, which makes the road to a longevity claim long.

Physics limits

  • Rapamycin's target does two jobsBlocking mTORC1 extends life in mice, but long use of rapamycin also blocks mTORC2, raising blood sugar and weakening immunity. Separating the two chemically is hard.
  • The energy sensor works in every tissueGenetic over-activation of AMPK thickens heart muscle, and one strong AMPK activator enlarged hearts in animals. Activators must be kept out of the heart or dosed gently.
  • Mouse lifespan gains may not carry overRapamycin extends mouse lifespan by roughly 10-25% depending on dose and sex, but mice age about 30 times faster than people and no human lifespan data exists.

An oral drug that switches on AMPK, the cell's low-fuel sensor, outside the brain, mimicking some effects of exercise and fasting.

mTOR

TORnado: a sharper rapamycin

Drugs that block only mTORC1, the part of the growth switch linked to longer life in animals, and spare mTORC2, which causes side effects.
